实验研究-益气健脾法对大鼠慢性溃疡性结肠炎NF-kB信号转导通路的实验研究

摘要:目的:益气健脾法对慢性溃疡性结肠炎大鼠结肠粘膜上皮组织NF一kB蛋白因子表达的影响,阐述补气健脾法对慢性溃疡性结肠炎的作用机制。方法:采用葡聚糖硫酸钠(dextron sulfate sodium,DSS)自由灌喂的方式建立UC的模型,用蒸馏水配制浓度为4%的DSS溶液,放到饮水瓶中自由饮用,连续用药两周,造模成功后分别给予益气健脾方中药煎剂高中低剂量组,西药美沙拉嗪组不同方法进行干预。结果:模型组大鼠结肠组织NF-kB蛋白和表达均明显高于正常组(P<0.01);健脾益肠散高、中剂量组结肠蛋白NF-kB蛋白表达均较模型组降低,差异有统计学意义(P<0.05);健脾益肠法低剂量组NF-kB蛋白表达亦低于模型组(P<0.05);健脾益肠法中剂量组对NF-kB的影响与美沙拉嗪组比较差异不显著,但明显优于低剂量组(P<0.05)。而健脾益肠法的高剂量组NF-kB蛋白表达明显低于中剂量组及西药组(P<0.05)。结论: 1、益气健脾法对溃疡性肠黏膜免疫的保护作用可能与结肠组织的蛋白NF-kB和基因表达有关2,益气健脾方治疗溃疡性结肠炎效果显著,优于西药组,值得临床推广应用。


Abstract: objective: to investigate the effect of invigorating spleen and qi on the expression of NF - kB protein in colonic epithelial tissues of rats with chronic ulcerative colitis, and to elucidate the mechanism of invigorating spleen and qi on chronic ulcerative colitis.Methods: using dextran sodium sulfate (dextron sulfate sodium, DSS) free filling and feeding way of UC model is set up, the concentration of 4% DSS solution with distilled water, into the water in the bottle drink freely, use two consecutive weeks, after the success of the building were given qi spleen method Chinese medicine medicinal broth including dose group, western medicine beauty salad oxazine groups of different intervention methods.Results: NF-kB protein and expression in colon tissues of the model group were significantly higher than those of the normal group (P < 0.01).The expression of colonic protein nf-kb in the high-dose and medium-dose groups was lower than that in the model group, and the difference was statistically significant (P < 0.05 or P < 0.01).The expression of NF-kB protein in the low dose group was lower than that in the model group (P < 0.05).The effect of the medium dose group on NF-kB was not significantly different from that of the mesalazine group, but was significantly better than that of the low dose group (P < 0.05).The expression of NF-kB protein in the high-dose group was significantly lower than that in the medium-dose group and the western medicine group (P < 0.05).Conclusion: 1. The protective effect of invigorating qi and strengthening spleen on UC intestinal mucosal immunity may be related to the protein nf-kb and gene expression in colon tissues. 2.


溃疡性结肠炎(ulcerative colitis,UC)又称慢性非特异性溃疡性结肠炎或特发性溃疡性结肠炎,是一种常见的慢性肠道疾病,发病部位主要在结肠的黏膜及黏膜下层,呈连续性弥漫性分布,也可累及直肠和乙状结肠[1]。主要发病人群是20-50岁之间,没有明显的性别差异。临床上主要以,腹痛、腹泻、里急后重、脓血便为主要的临床表现,本病轻重不等,缓解期与发作期交替进行,具有治愈率低,反复发作等特点,被世界卫生组织列为现代难治性疾病之一,其病因和发病机理以及对本病的治疗一直是学者们研究的热点[2].
